Antioxidant Potential of the Young Leave Methanolic Extract of Magnifera Indica in Alloxan Induced Diabetic Rat

Abstract: The study was designed to investigate the antioxidant potential of Magnifera indica young leave methanolic extract in alloxan induced diabetic rat. Albino rats each weighing 100-200g were given a peritoneal injection of 120mg of Alloxan per kg body weight. After 7 days the blood glucose level of the animals were checked to ascertain a diabetic state. Those that were diabetic were selected for the study. Significant increase (p<0.05) in lipid peroxidation and reduction in reduced glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) was observed in the serum, liver, kidney and heart of diabetic rats as compared to the normal control. Administering Magnifera indica young leave extract significantly reduced (p<0.05) lipid peroxidation and elevated the level of GSH, SOD, and CAT in serum, liver, kidney and heart of diabetic rats compared to untreated diabetic rats. These findings indicate that Magnifera indica young leave extract can reduce free radical mediated oxidative stress to cells in experimental diabetes mellitus.
Progression of nephropathy after islet of langerhans transplantation in alloxan-induced diabetic rats

Abstract: we studied the effects of islet of langerhans transplantation (it) on the kidney lesions of rats with alloxan-induced diabetes. forty-five inbred male lewis rats were randomly assigned to 3 experimental groups: group gl included 15 non-diabetic control rats (nc), group git included 15 alloxan-induced diabetic rats (dc), and group iii included 15 alloxan-induced diabetic rats that received pancreatic islet transplantation prepared by nonenzymatic method from normal donor lewis rats and injected into the portal vein (it). each group was further divided into 3 subgroups of 5 rats which were sacrificed at 1, 3, and 6 months of follow-up, respectively. clinical and laboratorial parameters were recorded in the mentioned periods in the 3 experimental groups. for histology, the kidneys of all rats of each subgroup were studied and 50 glomeruli and 50 tubules of each kidney were analyzed using light microscopy by two different investigators in a double blind study. the results showed progressive glomerular basement membrane thickening (gbmt), mesangial enlargement (me), and bowman's capsule thickening (bct) in the 3 experimental groups throughout the follow-up. these alterations were significantly more severe in dc rats at 6 months when compared to nc rats (p < 0.01). however, the degree of gbmt, me, and bct observed in dc rats was not statistically different from it rats at 1, 3, and 6 months. in addition, armanni-ebstein lesions of the tubules (ae) and tubular lumen protein (pro) observed in dc rats were also observed in it rats all over the study. these lesions were never present in nc rats. we conclude that it did not prevent progression of kidney lesions in alloxan-induced diabetic rats within 6 months after transplantation.
Anthocephalus cadamba (Roxb.) Miq., Rubiaceae, extract shows hypoglycemic effect and eases oxidative stress in alloxan-induced diabetic rats

Abstract: the hydroethanolic extract of the flowering tops of anthocephalus cadamba (roxb.) miq., rubiaceae, a bangladeshi medicinal plant, was studied for its potential hypoglycemic effect and antioxidant property in alloxan-induced diabetic rats. the extract induced significant reduction in serum glucose, and transaminases, e.g. aspartate transaminase (ast), alanine transaminase (alt) and alkaline phosphatases (alp), activities. significant changes in the thiobarbituric acid reactive substances (tbars), peroxidase and catalase levels during the experimental period were also observed. the results established that the hydroethanolic extract of the flowering tops of a. cadamba possesses hypoglycemic property and is able to protect liver and brain from oxidative damages caused by diabetes.
Antihyperglycemic activity of root of Berberis aristata D.C. in alloxan-induced diabetic rats

Abstract: The aim of this study is to examine the antihyperglycemic activity of root of Berberis aristata D.C. in alloxan-induced diabetic rats. Five groups of albino Wistar rats were used (n = 6). The two dose of 71.42 and 100 mg/kg body weight ethanol extract of B. aristata were selected for antidiabetic activity. Blood glucose levels were estimated in all the groups by the commercial kit (Span diagnostic Pvt. Ltd, Surat) on 1 st , 5 th , 10 th and 20 th day of the treatment with B. aristata. The serum cholesterol, triglycerides, HDL, liver glycogen and body weight were estimated on 20 th day of treatment in all the groups compared against diabetic control group. The different extracts of root of B. aristata were also tested for glucose tolerance test in normal fasted rats. The ethanol extract of root of B. aristata 71.42 and 100 mg/kg body weight showed a significant (P< 0.01) reduction of serum glucose level in alloxan induced diabetic rats at 15 th day as compared to diabetic control group. Cholesterol and triglycerides level were increased very significantly (P< 0.01), in diabetic animal when compared with normal control group. The level of cholesterol and triglycerides reduced very significantly (P< 0.01), when compared with diabetic control group. The level of HDL cholesterol was significantly (P< 0.05) increased in the extract treated group when compared to diabetic control group. In oral glucose tolerance test ethanol extract of B. aristata increase the glucose tolerance. It is concluded that the ethanol extract of B. aristata possess anti-diabetic activity in alloxan induced diabetic rats. The ethanol extract of B. aristata is very promising to develop standardized phytomedicine for diabetes mellitus.
ANTI-DIABETIC EFFECT OF MORUS ALBA ON RABBIT AS ANIMAL MODEL

Abstract: A study of ancient literature indicates that diabetes was fairly well known and well conceived as an entity in India. The nature has provided abundant plant wealth for all the living creatures, which possess medicinal virtues. Therefore, there is a necessity to explore their uses and to conduct Pharmacognostic and pharmacological studies to ascertain their therapeutic properties. In fact, nowadays diabetes is a global problem. Hence, the present study aims to open new avenues for the improvement of medicinal uses of Morus alba. for the area for diabetes. Another important objective of such study is to bring the anti-diabetic medicinal plants sector on a firm scientific footing, raise awareness and add value to the resource. Dried petroleum ether (60-80°C) extracts of leaves of Morus alba. were subjected for hypoglycemic activity in New Zealand rabbits (1.5-3.5 kg). Blood sugar level was determined using digital glucometer. The oral administration of leaf extracts at doses of 200 mg/ kg lead to a significant blood glucose reduction. This laid the foundation to study the active compounds of such anti-diabetic plants that are responsible for the hypoglycemic activities. It also proves the traditional claim of Kachh region with regard to Morus Alba for its anti-diabetic activity.
Antihyperglycemic activity of various fractions of Cassia auriculata linn. in alloxan diabetic rats

Abstract: Present work describes the potent antidiabetic fraction from flowers of Cassia auriculata Linn. Hydromethanolic extract along with its ethyl acetate and n-butanol fractions were evaluated for antidiabetic activity in alloxan-induced diabetes in rats. The n-butanol fraction exhibited significant reduction (p< 0.001) in blood glucose levels and was also found effective in restoring the blood lipids and proteins to normal level. The activity was found comparable with standard drug phenformin. The hydromethanolic extract and its fractions were subjected to preliminary qualitative chemical investigations which indicated the presence of phenolic compounds, carbohydrates, tannins, steroids and amino acids.

5. Evaluation of the hypoglycemic effect of aqueous extract of Phyllanthus amarus in alloxan-induced diabetic albino rats

Abstract: The hypoglycemic potential of aqueous extract of Phyllanthus amarus Schum was investigated in alloxan-induced diabetic albino rats. The extract at a dose of 260 mg/kg produced a significant (P< 0.05) reduction in blood glucose level by 112% at 24 h of oral administration. At 390 mg/kg, significant reduction (P< 0.05) in blood glucose levels of 102% (6 h) and 82% (24 h) were observed. A significant reduction (P<0.01) in blood glucose level of 81% and 61% (day 7) at doses of 130 and 260 mg/kg of extract respectively were observed. The extract also a highly significant (P<0.001) decrease in blood glucose level of 38% and 30% (day 14) at doses of 130 and 260 mg/kg respectively. On the administration of 390 mg/kg dose of extract, significant reduction (P<0.001) in blood glucose level of 41% on day 7 and 16% on day 14 were observed. The above results indicate the presence of hypoglycemic constituents in the plant, Phyllanthus amarus Schum.
Hypoglycemic Activity of Ethanolic Extract of Solanum nigrum Linn. Leaves on Alloxan Induced Diabetes Mellitus in Rats

Abstract: Diabetes is a metabolic disorder associated with hyperglycemia and caused by defect in insulin secrtion. Past few year some of the new bioactive drug isolated from plants showed anti- diabetic activity in clinical therapy . Hypoglycemic effect of these plant is due to their ability to restore the function of pancreatic β cells by causing an increase in insulin output or inhibit the intestinal absorption of glucose. Hence treatment with herbal drugs has an effect on protecting β cells and smoothing out fluctuation inglucose level. In present study we have screened alcoholic extracts of Solanum nigrum leaves for hypoglycemic effect in Albino rats. Different doses of alcoholic extract 50, 100, 200, 400 mg/kg of body weight were employed to evaluate alloxan induced diabetes with reference to standared Glibenclamide. Results indicated the alcoholic extract of leaves possesses significant hypoglycemic effect in dose dependent manner.
Methods to Reduce the Hypoglycemic Mortality of Alloxan in Diabetic Rabbit Model  [PDF]

Abstract: Objective: To explore an intervention method to reduce the mortality of alloxan diabetes model, and to preliminarily analyze the mechanism of alloxan induced animal death. Methods: Healthy New Zealand rabbits were randomly divided into injection group, control group, experimental group and blank group. The single injection group was injected with 100 mg/kg alloxan once. The control group was given 5% glucose solution and 100 mg/kg alloxan was injected in two times. The experimental group was given 5% glucose solution orally, 100 mg/kg alloxan, 7 mL 0.9% NaCl intravenously and 5 mL 5% glucose intraperitoneally immediately, and blood glucose was continuously monitored, 10 mL 5% glucose intravenously and 10 mL 5% glucose intraperitoneally every 4 h in the hypoglycemic stage. The blank group does nothing. Liver and kidney tissues at different time periods were stained with HE and organ index was evaluated. Results: 1) A single injection of 100 mg/kg alloxan without any intervention resulted in 100% mortality. Before modeling, oral administration of 5% glucose solution, divided into two injections of 100 mg/kg alloxan, mortality reached 100%; A single injection of 100 mg/kg alloxan and continuous intervention of normal saline and glucose for 20 h can significantly reduce the mortality of alloxan induced diabetic rabbit model. 2) Liver and kidney tissues were damaged in different degrees at different time periods, and liver and kidney indexes were significantly increased after alloxan injection compared with the normal group, with statistical significance (P > 0.05). Conclusion: 1) Every 4 hours of hypoglycemia, 10 ml 5% glucose was injected intravenously 10 ml 5% glucose intraperitoneally. It can reduce the death rate of alloxan diabetic rabbit model and shorten the time of blood glucose measurement. 2) After the injection of alloxan, acute lesions of liver and kidney may occur in different degrees, or one of the causes of acute death of experimental animals.
