Abstract:

目的:本研究旨在研究牛蒡提取物是否具有治疗非酒精性脂肪肝病(NAFLD)作用。方法:采用高脂饮食(HFD)喂食C57BL/6小鼠12周建立小鼠NAFLD模型,灌胃牛蒡提取物进行治疗。通过生化试剂盒检测小鼠血清中谷草转氨酶(AST)和谷丙转氨酶(ALT)的含量变化来观察牛蒡提取物对肝细胞损伤的影响。通过生化试剂盒检测小鼠血清中甘油三酯(TG)、血清总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)的含量变化来观察牛蒡提取物对脂质代谢的影响。用HE染色法观察牛蒡提取物对肝脏组织的损伤的影响。结果:HFD喂养小鼠12周后,小鼠发生了肝损伤,血清中AST和ALT的含量显著升高,HE染色结果也显示肝脏组织出现严重的病理损伤;并且也发生了脂质代谢紊乱,血清中TG、TC和LDL-C的含量显著升高。牛蒡提取物治疗可以显著降低NAFLD小鼠的肝损伤和脂质代谢紊乱,主要表现为:灌胃牛蒡提取物后,小鼠血清中的AST、ALT、TG、TC和LDL-C含量显著降低,并且肝脏组织病理损伤也得到显著的改善。结论:牛蒡提取物可以有效减轻HFD诱导的小鼠NAFLD。
Aim: To investigate whether Arctium lappa L. extract has therapeutic effects on non-alcoholic fatty liver disease (NAFLD). Methods: NAFLD model was induced in C57BL/6 mice by feeding with high-fat + high-sucrose diet for 12 weeks, and treated with Arctium lappa L. extract by gavage. To observe the impact of Arctium lappa L. extract on hepatocyte damage, biochemical kits were used to detect changes in aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in mouse serum. Similarly, to investigate the effect of Arctium lappa L. extract on lipid metabolism, biochemical kits were employed to measure alterations in triglyceride (TG), total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C) levels in mouse serum. The impact of Arctium lappa L. extract on liver tissue damage was assessed using hematoxylin and eosin (HE) staining. Results: After 12 weeks of high-fat diet (HFD) feeding, mice exhibited liver damage, with significant elevations in serum AST and ALT levels. HE staining results also indicated severe pathological damage to the liver tissue. Additionally, lipid metabolism disorder occurred, marked by significant increases in serum TG, TC, and LDL-C levels. Arctium lappa L. extract treatment significantly alleviated liver damage and lipid metabolism disorder in non-alcoholic fatty liver disease (NAFLD) mice, as evidenced by the notable reduction in serum AST, ALT, TG, TC, and LDL-C levels after gavage administration of Arctium lappa L. extract, accompanied by significant improvement in liver tissue pathological damage. Conclusion: Arctium lappa L. extract effectively mitigates HFD-induced NAFLD in mice.
