Abstract:

摘要 目的 探讨Snail在诱导食管癌细胞侵袭和迁移中的作用及机制。方法 采用慢病毒转染构建食管癌细胞株Eca-109-Snail和Eca-109-vc,Real-time PCR及Western blot实验分别检测其mRNA及蛋白表达水平,Transwell及细胞划痕实验检测细胞侵袭和迁移能力。 结果 慢病毒转染细胞株构建成功。经Real-time PCR及Western blot实验鉴定慢病毒转染后的Eca-109-Snail细胞Snail表达量升高,细胞表现出成纤维细胞样外形,细胞之间彼此分离;Eca-109-Snail细胞E-cadherin表达下降,Vimentin和MMP-2表达上升,其侵袭和迁移能力较空载体对照组增强,差异均具有统计学意义(P<0.01)。 结论 Snail通过诱导食管癌Eca-109细胞发生上皮间质转化,进而提高其侵袭和迁移能力
