Abstract:

摘要 目的 探讨FGF3和FGF10对人卵巢癌SKOV3细胞株迁移和侵袭的影响及其与Wnt/β-catenin信号通路的关系。方法 体外培养人卵巢癌SKOV3细胞,采用人工合成siRNA干扰细胞的FGF3和FGF10基因,运用Real-time PCR和Western blot分别从mRNA水平和蛋白水平检测干扰效果。Transwell检测FGF3和FGF10蛋白表达下调后细胞迁移和侵袭能力的变化。采用LiCl激活Wnt/β-catenin信号通路,运用Real-time PCR检测FGF3和FGF10基因的变化。结果 FGF3和FGF10基因沉默组细胞迁移和侵袭能力与阴性对照(NC)组细胞、未处理组SKOV3细胞相比明显减弱(P<0.05),NC组与未处理组SKOV3细胞迁移和侵袭能力差异无统计学意义(P>0.05)。 激活Wnt/β-catenin信号通路后,FGF3和FGF10基因水平增高。结论 FGF3和FGF10基因受Wnt/β-catenin信号通路的调控,促进人卵巢癌SKOV3细胞的迁移和侵袭
