Abstract:

摘要 目的 探讨长链非编码RNA LOC100294362在乳腺癌组织及细胞系中的表达情况,观察LOC100294362对乳腺癌MCF-7细胞增殖和侵袭的影响,并探索其可能的作用机制。方法 采用定量反转录PCR(qRT-PCR)方法检测乳腺癌组织及细胞系中LOC100294362的表达水平;MCF7细胞转染过表达或干扰LOC100294362后,采用MTT法和Transwell小室法检测上调或沉默LOC100294362对MCF-7细胞增殖和侵袭能力的影响,Western blot法检测p53蛋白表达。结果 与正常组织及细胞相比,LOC100294362在乳腺癌组织和细胞中显著高表达;过表达LOC100294362导致MCF-7细胞增殖和侵袭能力增加,p53蛋白表达降低;沉默LOC100294362导致MCF-7细胞增殖和侵袭能力受到抑制,p53蛋白表达升高。结论 LOC100294362可能通过抑制p53蛋白的表达从而促进乳腺癌细胞的增殖和侵袭
