峨眉拟单性木兰嫩茎诱导及再生培养研究

Abstract:

为奠定人工繁育和保护研究的基础,对峨眉拟单性木兰嫩茎诱导培养、不定芽的分化及组培苗再生进行了研究,建立起培养周期短、再生效率高的峨眉拟单性木兰组织培养体系。利用多因子正交实验设计及分析,筛选出最佳诱导培养基是MS + 2.0 mg/L 6-BA + 1.5 mg/L KT + 0.02 mg/L IAA。通过比较植物生长调节剂,发现最适的分化培养基为MS + 0.5 mg/L 6-BA + 0.02 mg/L NAA。最后,生根培养基以MS培养基添加NAA 0.5 mg/L,IBA 2.5 mg/L能有效地使再生苗产生根系。
Through the study of shoot induction culture, adventitious bud differentiation and plantlet rege-neration, a tissue culture system was established, which was short culture cycle and high regene-ration efficiency. The design and analysis of multivariate experiments showed, the best induction medium was MS + 2.0 mg/L 6-BA + 1.5 mg/L KT + 0.02 mg/L IAA. By comparing plant growth hor-mone, we found that the most suitable medium for differentiation was MS + 0.5 mg/L 6-BA + 0.02 mg/L NAA. Finally, MS medium with NAA 0.5 mg/L and IBA 2.5 mg/L can effectively produce roots of regenerated seedlings.
