Abstract:

采用真空渗入法,将萝卜 VPE1基因的干扰载体pRNAi- RsVPE1转入大白菜中,共收获2 414粒种子,在质量浓度为30 mg/L的卡那霉素筛选下获得抗性株297株,其中9株经PCR检测呈阳性,转化率为0.37%。Real-time PCR检测结果表明,pRNAi- RsVPE1的转入导致转基因大白菜叶片中同源的BraVPE在RNA水平上的表达量下降,验证了转基因植株的可靠性,为进一步深入研究BraVPE基因功能奠定了基础。
The vector pRNAi- RsVPE1 was transformed into Chinese cabbage by vacuum infiltration method and total 2 414 seeds were obtained. The kanamycin concentration 30 mg/L was determined to be the most suitable for selecting transgenic plants. 297 transgenic plants resistant to kanamycin were obtained,of which 9 positive transgenic plants were verified by PCR detection,and the transgenic rate was 0.37%. RT-PCR detection showed that the expression level of BraVPE in transgenic plants decreased significantly,reliability of transgenic plants was verified. Therefore ,the above results laid a foundation for further study in BraVPE gene functions.
