Abstract:
ABSTRACT: Objective To explore the function of Mycobacterium tuberculosis PPE36 protein in host-pathogen interactions. Methods The ppe36 gene was heterologously expressed in the nonpathogenic fast-growing Mycobacterium smegmatis MC2155 strain. And we used this recombinant strain to infect mouse macrophages. Then we analyzed the strain??s survival and cell death and change of cytokines within cells. Results PPE36 enhanced the intracellular survival of recombinant Mycobacterium smegmatis. Colonization of macrophages by MS-PPE36 was associated with cell death and significantly increased the expressions of TNF-α and IL-1β compared with controls. Conclusion Our study indicates that PPE36 delivered in a live Mycobacterium smegmatis vehicle can promote host cell apoptosis, modify the cytokines profile of macrophages, and enhance the persistence of mycobacterium within mouse macrophages
