%0 Journal Article %T 结核分枝杆菌PPE36蛋白增加耻垢分枝杆菌在小鼠巨噬细胞内生存及调节细胞因子的分泌<br>Mycobacterium tuberculosis PPE36 protein expressed in Mycobacterium smegmatis enhances mycobacterial survival and modulates cytokine secretion in mouse macrophages %A 梁丽娟 %A 米友军 %A 周明方 %A 王雪林 %J 西安交通大学学报(医学版) %D 2018 %R 10.7652/jdyxb201803016 %X 摘要:目的 研究结核分枝杆菌PPE36蛋白在宿主病原体相互作用时的功能。方法 将结核分枝杆菌PPE36蛋白异源表达于非致病性的耻垢分枝杆菌MC2155,利用该重组菌感染小鼠巨噬细胞,分析重组菌在细胞内的生存、细胞死亡、细胞因子的变化。结果 PPE36能够增加耻垢分枝杆菌在小鼠巨噬细胞内的生存能力,诱导细胞的坏死,增加TNF-α、IL-1β、减少IL-6的表达。结论 PPE36表达于耻垢分枝杆菌之后能够促进细胞死亡,影响细胞因子分泌,增加在小鼠巨噬细胞内的持留。<br>ABSTRACT: Objective To explore the function of Mycobacterium tuberculosis PPE36 protein in host-pathogen interactions. Methods The ppe36 gene was heterologously expressed in the nonpathogenic fast-growing Mycobacterium smegmatis MC2155 strain. And we used this recombinant strain to infect mouse macrophages. Then we analyzed the strain??s survival and cell death and change of cytokines within cells. Results PPE36 enhanced the intracellular survival of recombinant Mycobacterium smegmatis. Colonization of macrophages by MS-PPE36 was associated with cell death and significantly increased the expressions of TNF-α and IL-1β compared with controls.
