localizationandexpressionofslingshot-1linperipheraleosinophilsfrompatientswithacuteasthmaexacerbationKeywords: slingshot-1l,asthma,eosinophils,corticosteroids Abstract: abstract:backgroundeosinophilsplayapivotalroleinasthmaticairwayinflammation.wepreviouslyfoundasignificantlyhighexpressionofslingshot-1l(ssh-1l)inperipheraleosinophilsinacuteexacerbationsofasthma.objectivetoinvestigatetheexpressionandlocalizationpatternsofssh-1linperipheralbloodeosinophilsofasthmaticpatientsandtheirchangesaftertreatmentwithinhaledcorticosteroids.methodswerecruited4outpatientswithacuteexacerbationsofasthmawhoreceivednopreviouscorticosteroidtreatmentand1healthyvolunteer.fromallthesubjects30mlperipheralvenousbloodsampleswerecollectedbeforeandaftera3-monthtreatmentwithinhaledfluticasone.theeosinophilswereisolated,purifiedandcounted,andtheexpressionsofssh-1lintheeosinophilswereexaminedbyrt-pcrandwesternblotting.thelocalizationofssh-1lphosphatasesintheperipheraleosinophilswasdetectedbyimmunofluorescenceassayinonepatient.resultsssh-1lphosphatasesdistributeddiffuselyinthecytoplasm,especiallydensenearthemembraneoftheperipheraleosinophils.glucocorticoidstreatmentresultedinasignificantreductioninboththessh-1lmrnaexpression(0.7403±0.1124vs0.4101±0.0363,p=0.001)andssh-1lproteinexpression(0.3410±0.1337vs0.1543±0.0551,p=0.039).conclusionsahighexpressionofssh-1linperipheraleosinophilsinacuteexacerbationsofasthmamayplayaroleintheactivationandmigrationofeosinophils.theefficacyofinhaledcorticosteroidsinasthmacontrolmightbepartlyattributedtoadown-regulatedexpressionofssh-1l.
