%0 Journal Article %T localizationandexpressionofslingshot-1linperipheraleosinophilsfrompatientswithacuteasthmaexacerbation %A 张卫珍 %A 赵海金 %A 邹飞 %A 李文军 %A 蔡绍曦 %J 南方医科大学学报 %D 2011 %X abstract:backgroundeosinophilsplayapivotalroleinasthmaticairwayinflammation.wepreviouslyfoundasignificantlyhighexpressionofslingshot-1l(ssh-1l)inperipheraleosinophilsinacuteexacerbationsofasthma.objectivetoinvestigatetheexpressionandlocalizationpatternsofssh-1linperipheralbloodeosinophilsofasthmaticpatientsandtheirchangesaftertreatmentwithinhaledcorticosteroids.methodswerecruited4outpatientswithacuteexacerbationsofasthmawhoreceivednopreviouscorticosteroidtreatmentand1healthyvolunteer.fromallthesubjects30mlperipheralvenousbloodsampleswerecollectedbeforeandaftera3-monthtreatmentwithinhaledfluticasone.theeosinophilswereisolated,purifiedandcounted,andtheexpressionsofssh-1lintheeosinophilswereexaminedbyrt-pcrandwesternblotting.thelocalizationofssh-1lphosphatasesintheperipheraleosinophilswasdetectedbyimmunofluorescenceassayinonepatient.resultsssh-1lphosphatasesdistributeddiffuselyinthecytoplasm,especiallydensenearthemembraneoftheperipheraleosinophils.glucocorticoidstreatmentresultedinasignificantreductioninboththessh-1lmrnaexpression(0.7403±0.1124vs0.4101±0.0363,p=0.001)andssh-1lproteinexpression(0.3410±0.1337vs0.1543±0.0551,p=0.039).conclusionsahighexpressionofssh-1linperipheraleosinophilsinacuteexacerbationsofasthmamayplayaroleintheactivationandmigrationofeosinophils.theefficacyofinhaledcorticosteroidsinasthmacontrolmightbepartlyattributedtoadown-regulatedexpressionofssh-1l. %K slingshot-1l %K asthma %K eosinophils %K corticosteroids %U http://www.j-smu.com/oa/darticle.aspx?type=view&id=201106928
