endogenousneuregulin-1expressionintheanteriorpituitaryoffemalewistar-furthratsduringtheestrouscycleKeywords: neuregulin-1,estrouscycle,anteriorpituitary,phospho-erbb-4,wistar-furthrats Abstract: abstract:objectivetostudythechangesinendogenousneuregulin-1(nrg1)expressionintheanteriorpituitaryoffemalewistar-furthratsindifferentphasesoftheestrouscycle.methodsfemalewistar-furthratsduringestrouscycleswereused.rt-pcrwasemployedtostudythechangesintheexpressionofnrg1isoformsandtheircognatereceptorserbb-2anderbb-4intheanteriorpituitaryindifferentphasesoftheestrouscycle.westernblottingwasusedtodetectnrg1expressionattheproteinlevel.immunofluorescencestainingwasusedtoidentifyhypophysealcellsexpressingnrg1andobservethelocalizationanddistributionofnrg1andfunctionalphosphorylationoferbb-4.theco-expressionofnrg1anderbb-4intheanteriorpituitaryofrhesusmonkeywasalsoinvestigated.resultssomeofthenrg1isoforms,especiallytypeiiinrg1s,wereexpressedatahigherlevelduringtheestrouscyclei(e1)andestrouscycleii(e2),aresultconsistentwiththatofwesternblottingforsamplesoftheanteriorpituitariescollectedatthesephases.immunofluorescencestainingidentifiedthegonadotrophsasthemainsourceofnrg1,andshowedanextensivedistributionofnrg1intheanteriorpituitaryine1ande2phasesaccompaniedbyapparentphosphorylatedactivationoferbb-4.adjacentdistributionofnrg1-anderbb-4-positivecellswasalsoobservedintheanteriorpituitaryofmalerhesusmonkeys.conclusionourresultsprovideevidencefortheexpressionofmultiplenrg1isoformsandthepresenceofnrg1/erbb-4signalingintheanteriorpituitaryoffemalewistar-furthrats.thissignalingdemonstratesanestrouscyclephase-relatedpattern.additionally,nrg1/erbb-4-basedjuxtacrinesignalingmayexistintheanteriorpituitaryofmalenon-humanprimate.
