%0 Journal Article %T endogenousneuregulin-1expressionintheanteriorpituitaryoffemalewistar-furthratsduringtheestrouscycle %A 赵炜疆 %A 任颂光 %J 南方医科大学学报 %D 2011 %X abstract:objectivetostudythechangesinendogenousneuregulin-1(nrg1)expressionintheanteriorpituitaryoffemalewistar-furthratsindifferentphasesoftheestrouscycle.methodsfemalewistar-furthratsduringestrouscycleswereused.rt-pcrwasemployedtostudythechangesintheexpressionofnrg1isoformsandtheircognatereceptorserbb-2anderbb-4intheanteriorpituitaryindifferentphasesoftheestrouscycle.westernblottingwasusedtodetectnrg1expressionattheproteinlevel.immunofluorescencestainingwasusedtoidentifyhypophysealcellsexpressingnrg1andobservethelocalizationanddistributionofnrg1andfunctionalphosphorylationoferbb-4.theco-expressionofnrg1anderbb-4intheanteriorpituitaryofrhesusmonkeywasalsoinvestigated.resultssomeofthenrg1isoforms,especiallytypeiiinrg1s,wereexpressedatahigherlevelduringtheestrouscyclei(e1)andestrouscycleii(e2),aresultconsistentwiththatofwesternblottingforsamplesoftheanteriorpituitariescollectedatthesephases.immunofluorescencestainingidentifiedthegonadotrophsasthemainsourceofnrg1,andshowedanextensivedistributionofnrg1intheanteriorpituitaryine1ande2phasesaccompaniedbyapparentphosphorylatedactivationoferbb-4.adjacentdistributionofnrg1-anderbb-4-positivecellswasalsoobservedintheanteriorpituitaryofmalerhesusmonkeys.conclusionourresultsprovideevidencefortheexpressionofmultiplenrg1isoformsandthepresenceofnrg1/erbb-4signalingintheanteriorpituitaryoffemalewistar-furthrats.thissignalingdemonstratesanestrouscyclephase-relatedpattern.additionally,nrg1/erbb-4-basedjuxtacrinesignalingmayexistintheanteriorpituitaryofmalenon-humanprimate. %K neuregulin-1 %K estrouscycle %K anteriorpituitary %K phospho-erbb-4 %K wistar-furthrats %U http://www.j-smu.com/oa/darticle.aspx?type=view&id=201106921
