鼠肝炎冠状病毒非结构蛋白1内保守序列LLRKxGxKG的功能研究

Abstract:

目的?研究鼠肝炎冠状病毒(MHV)非结构蛋白1(NSP1)内的保守氨基酸序列LLRKxGxKG的功能。方法??扩增并构建MHVNSP1正常基因及LLRKxGxKG序列缺失突变的NSP1基因,分别将其插入真核表达载体pcDNA3.1中,获得真核表达质粒。用所构建的真核表达质粒转染L929细胞,经新城疫病毒(NDV)刺激诱导后,采用ELISA法检测IFN-β的表达水平,观察NSP1正常或突变蛋白对IFN-β表达的影响。通过与含CAT和Luc报告基因的重组质粒进行共转染,观察NSP1正常或突变蛋白对IFN-β启动子活性和干扰素刺激应答元件(ISRE)活性的影响。通过与三种报告基因(pRLuc-CMV、pGL3-basic、pGL3-control)共转染,观察NSP1正常或突变蛋白对共转染基因表达的影响。结果??MHVNSP1对真核细胞产生干扰素有一定的影响,并可显著抑制IFN-β启动子或ISRE应答元件的活性,且该抑制作用不受LLRKxGxKG序列缺失的影响。同时,MHVNSP1还可强烈抑制多种共转染报告基因的表达。该抑制作用在LLRKxGxKG序列缺失后显著降低。结论??MHVNSP1对Ⅰ型干扰素信号通路有特异的抑制作用。?LLRKxGxKG序列是MHVNSP1的一个重要功能域,在抑制共转染基因表达过程中发挥着重要作用。
