硒对黄曲霉毒素B1暴露肉鸡肝脏氧化损伤的保护作用

Abstract:

本试验旨在研究硒(Se)对黄曲霉毒素B1(AFB1)毒性的缓解作用及其可能机制。选取1日龄爱拔益加(AA)肉仔鸡90只,随机分为3组(每组3个重复,每个重复10只):基础饲粮组(对照组)、基础饲粮+100μg/kgAFB1组(AFB1组)、基础饲粮+100μg/kgAFB1+0.3mg/kgSe组(Se组),试验期为42d。结果表明:1)与对照组相比,AFB1组肉鸡肝脏重显著降低(P<0.05);血清谷丙转氨酶活性显著升高(P<0.05);肝脏丙二醛和过氧化氢含量显著升高(P<0.05),总超氧化物歧化酶活性、谷胱甘肽含量和谷胱甘肽过氧物酶活性则显著降低(P<0.05);肝脏组织病理切片观察到多处出血位点及肝细胞坏死和萎缩;免疫组织化学和蛋白质印迹分析发现核转录因子E2相关因子2(Nrf2)蛋白在肝细胞核内表达显著增加(P<0.05),凋亡因子含半胱氨酸的天冬氨酸蛋白水解酶-3(caspase-3)在AFB1组胞核内表达。2)在100μg/kgAFB1饲粮中加入Se能在一定程度上缓解AFB1引起的肉鸡肝功能紊乱和肝脏氧化应激损伤,并使肝细胞胞核Nrf2表达水平显著下降(P<0.05)。由此可见,Se能够缓解AFB1引起的肉鸡肝脏功能紊乱和氧化损伤,增加肉鸡抗氧化能力。
