PlantsregenerationfromprotoplastandtheirgeneticvariationinfoxtailmilletDOI: 10.7668/hbnxb.1993.S2.006, PP. 35-39 Keywords: foxtailmillet(SetariaitalicaBeau.),protoplast,plantregeneration,geneticvaria-tion Abstract: Compactcalliderivedfromimmaturespikeletofafoxtailmilletvariety—Jigu11cann'tbedirectlyusedforprotoplastisolationbecauseofitsfirmphysicalstructure,andmustbeloosenedwithsubculturinginM1,M2andM3mediasuccessivelyandalteringthesemediacompo-sitions.Theloosenedcallicanbeselectedfromtheregulationandusedforprotoplastisolationsuccessfully.RateofprotoplastdivisioninKM8Pmediumwas12.3—33.5%.Calliderivedthroughprotoplastdivisionarelooseandcann'tbeuseddirectlyforplanregenerationbecauseofitssoftphysicalstructure.WhentheyweresubculturedinN6—1,N6—2,N6—3andN6—4media,inwhichthemediacompositionswerechanged,thecompactcalliwereobtainedand129plantletswereregeneratedfromthem.101plants,whichgrewtomaturityaftertransplantingtheplantletsintofield,exhibitedsterilityinsomedegree.Mostofthesubsequentlinesderivedfromtheregen-eratedplantsweresterileandonlytwolinescouldgetnormalreproduction.
