IdentificationofAnamorphYeastofTremellaaurantialbaandOptimizationofMediumCompositionforProductionofExopolysaccharidesKeywords: Tremellaaurantialba,anamorphyeast,largesubunitribosomalRNAgene,internaltranscribedspacer,exopolysaccharides,centralcompositedesign Abstract: Ayeast-likefungusstrainB1isolatedfromwildfungusTremellaaurantialbawasidentifiedandinitiallycharacterized.TwophylogenetictreesweregeneratedbasedonthesequencesoflargesubunitribosomalRNAgeneD1/D2regionsandinternaltranscribedspacer(ITS)regionsofrelatedfungi,respectively.TheanalysisofD1/D2regionsandITSsequencesshowedthatfungusB1wasclusteredtogetherwithT.aurantialba,T.aurantiaandT.microsporeinthephylogenetictrees.BoththemorphologicalcharacteristicandphylogeneticanalysisestablishedthatfungusB1wasoneoftheanamorphstrainsofT.aurantialbaandbelongstoTremellagenus.Afermentationmediumforexopolysaccharides(EPS)productionbyT.aurantialbaB1.Plackett-Burmendesignwasusedtoevaluatetheeffectsofdifferentcomponentsintheculturemedium.GlucoseandyeastextracthavesignificantinfluenceontheEPSproduction.Theconcentrationsoftwofactorswereoptimizedsubsequentlyusingcentralcompositedesignandresponsesurfaceanalysis.Theresultsshowedthat49.2g/Lglucoseand10.4g/LyeastextractcouldleadtothemaximumproductionofEPS(4.99g/L).Theoptimizedmediumledtoa1.5-foldenhancementoftheproductionofEPSbyT.aurantialbaB1,ascomparedwiththatwithoutoptimization.
