%0 Journal Article %T IdentificationofAnamorphYeastofTremellaaurantialbaandOptimizationofMediumCompositionforProductionofExopolysaccharides %A 刘春卉 %A 杨秀伟 %A 俞建国 %A 马维新 %A 瞿伟菁 %A 包艳洁 %A 武文斌 %A 庄秀园 %J 过程工程学报 %D 2012 %X Ayeast-likefungusstrainB1isolatedfromwildfungusTremellaaurantialbawasidentifiedandinitiallycharacterized.TwophylogenetictreesweregeneratedbasedonthesequencesoflargesubunitribosomalRNAgeneD1/D2regionsandinternaltranscribedspacer(ITS)regionsofrelatedfungi,respectively.TheanalysisofD1/D2regionsandITSsequencesshowedthatfungusB1wasclusteredtogetherwithT.aurantialba,T.aurantiaandT.microsporeinthephylogenetictrees.BoththemorphologicalcharacteristicandphylogeneticanalysisestablishedthatfungusB1wasoneoftheanamorphstrainsofT.aurantialbaandbelongstoTremellagenus.Afermentationmediumforexopolysaccharides(EPS)productionbyT.aurantialbaB1.Plackett-Burmendesignwasusedtoevaluatetheeffectsofdifferentcomponentsintheculturemedium.GlucoseandyeastextracthavesignificantinfluenceontheEPSproduction.Theconcentrationsoftwofactorswereoptimizedsubsequentlyusingcentralcompositedesignandresponsesurfaceanalysis.Theresultsshowedthat49.2g/Lglucoseand10.4g/LyeastextractcouldleadtothemaximumproductionofEPS(4.99g/L).Theoptimizedmediumledtoa1.5-foldenhancementoftheproductionofEPSbyT.aurantialbaB1,ascomparedwiththatwithoutoptimization. %K Tremellaaurantialba %K anamorphyeast %K largesubunitribosomalRNAgene %K internaltranscribedspacer %K exopolysaccharides %K centralcompositedesign %U http://www.jproeng.com/qikan/Cpaper/zhaiyao.asp?bsid=16708
