MetabolismKineticsofGlucoseinAnchorage-dependentCellCulturesKeywords: animalcellculture,metabolism,kinetics,glucose,maintenanceenergy Abstract: ThekineticmodelofglucosemetabolismwasestablishedandsuccessfullyappliedtobatchculturesofrCHOandrBHKcells.Itwasfoundthatalargeamountofglucosewasutilizedforcellmaintenance,andtheoverwhelmingmajorityofmaintenanceenergyfromglucosewasbyitsanaerobicmetabolisminbothrBHKandrCHOcellcultures.Theoverallmaintenancecoefficientsfromaerobicmetabolismwere1.9?10-13mmol/(cell？h)forrCHOcellsand7?10-13mmol/(cell？h)forrBHKcells.Inaddition,allGO/TandEO/TgraduallyincreasedwiththesametrendasthecellgrowthinthecultureofbothrCHOandrBHKcells.Theoverallmoleculeyieldcoefficientsoflactatetoglucosewere1.61forrCHOcellsand1.38forrBHKcells.Theyieldcoefficientsofcelltoglucosewere4.5?108cells/mmolforrCHOcellsand1.9?108cells/mmolforrBHKcells,respectively.
