Abstract:

Masusalmon,Oncorhynchusmasoumasou,isoneofthemostvaluablefisheryspeciesthathasbeenintroducedtoChina,thoughtodatenostudiesonthegeneticdiversityandgeneticrelationshipamonghatcherypopulationshasbeenperformedwithmolecularmarkers.Weundertooksuchastudyandsampled120individualsfromthreehatcherystocksandanalyzed20microsatelliteloci.Alllociwerepolymorphicandatotalof91allelesweredetected.Arelativelylowlevelofgeneticdiversitywasrevealedwitheffectivenumberofalleleof3.1094,3.3299and3.1894andexpectedheterozygosityof0.6600,0.6648and0.6638inthethreestocks,respectively.DeviationsfromHardy-Weinbergequilibriumwerefoundduetoheterozygotedeficit.Accordingly,evidenceofgeneticbottleneckswerefoundinthethreestocks.Anindividualassignmenttestdemonstratedthat85%ofindividualswerecorrectlyassignedintotheiroriginalstocks.PairwiseFstrevealedthatsignificantdifferentiationoccurredbetweenthesethreestocks.Theresultsofthestudyindicatedthatdisequilibriumofgeneticstructureanddifferentiationhasoccurredinallthreestocks.ThisinformationcollectivelyprovidesabasisformeasurestoavoidoflossofgeneticdiversityandintrogressioninChineseaquaculture.
