Abstract:

Foodrestriction(FR)andrefeeding(Re)havebeensuggestedtoimpairbodymassregulationandtherebymakingiteasiertoregainthelostweightanddevelopover-weightwhenFRends.However,itisunclearifthisisthecaseinsmallmammalsshowingseasonalforgingbehaviors.Inthepresentstudy,energybudget,bodyfatandserumleptinlevelweremeasuredinstripedhamstersthatwereexposedtoFR-Re.Theeffectsofleptinonfoodintake,bodyfatandgenesexpressionsofseveralhypothalamusneuropeptidesweredetermined.Bodymass,fatcontentandserumleptinleveldecreasedduringFRandthenincreasedduringRe.LeptinsupplementsignificantlyattenuatedtheincreaseinfoodintakeduringRe,decreasedgenesexpressionsofneuropepetideY(NPY)andagouti-relatedprotein(AgRP)ofhypothalamusandleptinofwhiteadiposetissue(WAT).Hormone-sensitivelipase(HSL)geneexpressionofWATincreasedinleptin-treatedhamstersthatwerefedadlibitum,butdecreasedinFR-Rehamsters.ThisindicatesthattheadaptiveregulationofWATHSLgeneexpressionmaybeinvolvedinthemobilizationoffatstorageduringRe,whichpartlycontributestotheresistancetoFR-Re-inducedoverweight.LeptinmaybeinvolvedinthedownregulationsofhypothalamusorexigenicpeptidesgeneexpressionandconsequentlyplaysacrucialroleincontrollingfoodintakewhenFRends.
