SequencingandphylogeneticanalysisofthePyrgilaudaruficollis(Aves,Passeridae)completemitochondrialgenomeDOI: 10.11813/j.issn.0254-5853.2014.2.081, PP. 81-91 Keywords: Pyrgilaudaruficollis,Completemitochondrialgenome,Phylogeny Abstract: Inthisstudy,bothlongPCRandconservedprimerswalkingsequencingmethodswereusedtodeterminethecompletesequenceoftheofPyrgilaudaruficollismitochondrialgenome(KC836121).TheresultsshowedthatthecompletemitochondrialgenomeofP.ruficollisis16909bpinlengthwith55.0%A+Tcontent,harboringthetypical37genes.ThemitogenomehadthesamegeneorderwiththatofPodoceshendersoni.AllproteincodinggenesstartedwithATGcodon,exceptND3withGTG.Forthestopcodonusage,mostgenesterminatewithcodonsTAAorTAG,butND5terminatedwithAGA,whileND1andCOIgeneswithAGG,andboththegenesCOⅢandND4haveanincompleteterminationcodon(T).Thesecondarystructuresof22tRNAgeneswerealsopredicted,showingthatalltRNAscanformtypicalclover-leafsecondarystructures,exceptforthetRNASer(AGN)whichlosestheDHUarm,whiletRNAPheharboranextranucleotideinsertedintheTψCarm.Thepredictedsecondarystructuresof12SrRNAand16SrRNAexhibit47helicesin4domainsand60helicesin6domainsrespectively.ThecontrolregionofP.ruficolliswiththelengthof1305bpwaslocatedbetweentRNAGluandtRNAPhe,andtypicaldomainsofwhichcouldbefoundasotherbirdgroups.Usingthedatafrom13mitochondrialprotein-codinggenes,resultsofafinalphylogeneticanalysisstronglysupportsthetraditionalviewthatP.ruficollisiscloselyrelatedwithPasseridaeandFringillidae.
