大豆凝集素含量测定方法的改进与种质资源分析

Abstract:

大豆凝集素是大豆中一种主要的抗营养因子,对动物生长有一定的抑制作用,其含量作为评定动物饲料品质的重要指标。本文对血凝法测定大豆凝集素含量的方法进行了改进,并利用该方法对64份种质资源进行了凝集素含量的分析。结果表明:整粒种子粉碎过40目筛、样品与60~90℃石油醚比(W∶V)1∶10脱脂10h、样品与生理盐水(W∶V)1∶10抽提10h为宜,反应pH值在5.0~9.0为宜,反应温度在25℃左右为血凝法的最适条件,该方法具有快速鉴定、经济实用、适合大规模资源筛选等特点。64份种质资源中凝集素含量最低为1.88mgg-1,最高为37.5mgg-1,平均为11.91mgg-1,85%的品种(系)在1.88mgg-1到12.06mgg-1之间。
