甜玉米脂氧合酶酶学特性及热失活动力学

Abstract:

采用分光光度法测定甜玉米脂氧合酶(LOX)的酶学特性及热失活动力学性质,考察了温度、pH值、金属离子及其络合剂浓度对脂氧合酶活性的影响,同时测定了LOX酶促反应动力学及热失活动力学参数,结果表明:脂氧合酶的最适温度为30℃,最适pH值为7.5,在pH值为6的环境中,LOX活性保持相对稳定;Ca2+、K+、Mn2+对LOX活性具有一定的激活作用;而Fe3+、Cu2+、金属离子络合剂及巯基试剂对LOX活性具有抑制作用;LOX催化反应动力学符合米氏方程所描述的单底物酶促反应动力学,其Km为4.37mmol·L-1,Vmax为13.00U·min-1;LOX的热失活遵循一级反应动力学规律,反应活化能为116.39±0.52kJ·mol-1,该试验为甜玉米加工贮藏中的LOX活性控制提供了关键参数。
