Abstract:

En el presente estudio se llevó a cabo la detección de antígenos predominantes de Trichinella spiralis en 3 modelos experimentales (Rata, ratón y conejo), mediante Western blot (WB) autorradiografía. A los animales experimentales se les administró larvas infectantes del parásito y se sangraron pre-inoculación y a la cuarta semana postinfección. El AST (Antígeno soluble de Trichinella) se obtuvo a partir de larvas de músculo de rata experimentalmente infectadas. Con el AST se realizaron corridas electroforé-ticas en condiciones reductoras, los geles no te idos fueron transferidos a papel de nitrocelulosa (NC), los anticuerpos unidos a papel de NC se marcaron con Proteína A de Staphylococcus aureus (SPA) y las bandas inmunorreactivas se observaron por autorradiografía. En WB se encontró que los animales de las tres especies, infectados con T. spiralis reconocen varios componentes antigénicos con rango de peso molecular de 29-97 kDa con predominio de un triplete de 50 kDa, 47 kDa y 45 kDa en el 100% de los sueros estudiados In this study, the detection of dominant antigens of Trichinella spiralis in three experimenta modeis (rat, mouse and rabbit), was carried out by Western Blot autoradiography (WB). Healthy animals were considered as controls. Experimenta animals were infected with infective larvae of T. spiralis: Animals were bleed before inoculation and at the fourth week post infection. The tri-chinella soluble antigens (AST) were obtained from muscles of experimentally infected rats. The AST were separated by reducing electrophoresis. Proteins in not stained gels were transferred to nitrocellulose paper sheets (NC), antibodies linked to NC were labeled with Staphylococcus aureus protein A SPA y (SPA) and the immune reactive bands were observed by autoradiography. By WB, T. spiralis infected animals recognized several antigenic componentes with a molecular weight ranging from 29 to 97 kDa, with dominant bands at 50, 47 and 45 kDa observed in 100% of studied sera.
