The Quantitative Method of Pravastatin in Plasma by Gas Chromatograoahy/Mass Spectrometry

Abstract:
Pravastatin sodium is a cholesterol-lowering agent and potent inhibitor of 3hydroxy-3-methylglutaryl coenzyme A (HMGCoA) reductase. The plasma concentration is very low. This paper reports a highly sensitive and quantitative method specific for determination of pravastatin in human plasma based on the selective ion monitoring (SIM) mode in gas chromatography/electron impact ionization mass spectrometry. The plasma samples were acidified to pH 4. 0 and then extracted by ethyl acetate. The plasma concentrations of pravastatin in healthy volunteers were detected by selecting m/z 462 as quantitation ion after the sample was methyl esterification with diazomethane and silanization with BSTFA. The minimun detectable limit was 0. 015ng. The standard curve was linear between 1.0 to 60 ng/mL, with a correlation coefficient more than 0. 99. The coefficiens of variation was less than 7.7%. The recovery can reach 90%. This method has been successfully used for the study of pharmacokinetics of pravastatin.
