Enantiomeric Resolution by Chiral Ligand-Exchange Chromatography with Chiral Mobile Phase

Abstract:
Chiral ligand-exchange chromatography(CLEC),in which three types of chiral LC systems are generally employed,is revieved with emphasis on the chiral mobile phase. The main points include : (A) Mechanism of chiral ligand-exchange chromatography. Equations are given describing the retention and separation selectivity of two enantiomeric species in a chromatographic system containing a chiral selector in the stationary and/or in the mobile phase. It is shown that the total column enantioselectivity generally differs from the enantioselectivity of the selector-selectand interaction in solution. (B)Parameters affecting ligand exchange. Twelve factors are disscused including metal ions, metal/ligand ratio, concentration of metal/complex, stationary phase, eluent PH, elution order,organic medifier, ion-pair reagent, ion strength of mobile phase, temperature,stereoselectivity, and mutual recognition. (C) Applications. Some enantiomers separated by CMPs of CLEC are presented as examples.
