A METHOD OF DETECTION FOR SUGAR AND ANTIGEN SITES ON HYPHAE SURFACES OF WHEAT STEM RUST BY PRE-EMBEDDING CYTOCHEMICAL TECHNIQUE

Abstract:
This paper described a method of detection for sugar and antigen sites on hyphae surfaces of wheat stem rust by pre-embedding cytochemical technique.After rust- infected wheat leave had been pre-fixed with 1% glutaraldehyde and 4% paraformallehyde,the sliced leaf samples were incubated with 2 lectin probes,or 2 antibodies and protein A-gold res- pectively,then following by post-fixation with 2% OsO_4,dehydration in a graded ethanol se- ries and embedding in Epon-Aratdite.It was observed that 2 kinds of sugar sides on the sur- faces of rust hyphae located between host cells had been accurately detected by 2 lectin probes, and 2 kinds of antigenic sites on the hyphae surfaces had been revealed by 2 antibodies and pro- tein A-gold.The result here indicates obviously that this method is suitable for detection of components on surfaces of rust hyphae located between host cells.
