RECOMBINATION AND EXPRESSION OF PATERNAL GENE OF ISOZYMES IN GYNOGENETIC OLIVE FLOUNDER PARALICHTHYS OLIVACEUS

Abstract:
Gynogenetic induction of olive flounder Paralichthys olivaceus was carried out by cold shock after activation of the eggs with sperm which had been inactivated with UV irradiation. Biochemical samples of 57 gynogenetic flounder (in a family) were collected in May, 1999 8 isozymes, including LDH, IDHP, PGM, ACP, ADH, SDH, GDH, CAT, were analyzed by starch gel electrophoresis. These isozymes were polymorphic in wild population. Two loci, Ldh C and Cat had recombination in gynogenetic population. The recombination rates between isozyme locus and centromere were 52 6% and 29 8%, respectively. Paternal gene of Idhp 1 was found expressing at a low level in gynogenetic progenies. Mean proportion of polymorphic loci and the average heterozygosities of gynogenetic population were 7 0% and 0 035, respectively, much lower than that of wild population. The result suggests that gynogenesis involving retention of the second polar body is not a good method for making pure lines, and gynogenesis involving suppression of the first cell division may be feasible.
