A nonapeptide library was constructed by cloning randomly synthesizedoligonucleotides into ptacG4. Millions of different nonapeptides can be expressed inE. coli cell in a loop of GAL4 zinc finger structure, which was derived from the plasmidptacG4. Compared with the surface displayed peptide library, cytoplasmic library can bescreened by biological activity and can be easily kept in expression plasmid.
