CLONING AND IDENTIFICATION OF fliC~i GENE OF SALMONELLA TYPHIMURIUM
鼠伤寒沙门氏菌Ⅰ相鞭毛蛋白基因fliC~I的克隆和鉴定

Purified Salmonella typhimurium 8705 chromosomal DNA was digested with EcoR I. DNA fragments above 400bp were obtained by Sepharcyl S-400 chromatography and cloned into vector pGEM-3Zf (-) , then transformed into host cell LC2a(hag-is recA-). One out of 6013 transformants was found to be motile and this clone was named pGI4015. Miniprep proved that pG I 4015 contained an inserted fragment about 15.3kb. Motility / inhibition tests as well as Southern blot hybridization showed that pG I 4015 bear the H-li gene of S. typhimurium. With the aid of BamH I and Sal I site, sequences unrelated to flagellin in the cloned DNA fragment was removed, and a 3.8kb fragment containing fliC gene was subcloned.
