RESTRICTION PATTERNS AND NUCLEOTIDE SEQUENCE OF THE POLYHEDRIN GENE OF SPODOPTERA LITURA NUCLEAR POLYHEDROSIS VIRUS

Abstract:
Digestion of Spodoptera litura nuclear polyhedrosis virus (SINPV Guangzhou isolate) with restrictionendonucleases EcoRI.XbaI.Xhol.Bam HI Pst I SacI .Hind and SmaI resulted in 26.26.24.20. 13. 17.9 and 1fragments, respectively. The average size of the genome is about 136.OKbp. The polyhedrin gene of SINPV waslocated on the Xba I O fragments by southem hybridization methed using a DIG labeled AcMNPV polyhedringene as probe. With a coding sequence of 780 nucleotides, corresponding to a protein of 249 amino acid, thepolyhedrin gene of SINPV is the longest polyhedrin gene known to date together with the Spodoptera littoralisNPV polyhedrin gene. This gene codes for a putative protein with an Mr 29236 and the predicted amino acidsequence shows high homology with those of other NPVs.
