THE EFFECT OF THE ANTIC ANCER DRUG CIS-DDP ON THE DENATURATION OF DNA

Abstract:
Cis-diamminedchloroplatinum( II ) (cis-DDP) is a kind of antitumous medicine. It bonds to the N-7 of the guanine of DNA and causes intrastrand or intrastrand cross - linking. Our experiments induce fluorecence techniques to analyze the influence of cis-DDP on DNA denaturation so as to give some information on the interaction between cis-DDP and DNA. The results are as fellows:(1) proved that as - DDP made hydrogen bond of G - C easy to be damaged in vitro.(2) Got the percent of damaged hydrogen bonding in DNA at different temperature befor and after cis-DDP was added(3) the influence of the percent of G-C base in DNA.(4) the influence of the concentration of cis-DDP(5) the brief discussion of the mechanism of antitumor
