TWO-DIMENSIONAL NUCLEAR MAGNETIC RESONANCE INVESTIGATION OF GRAMICIDIN S IN REVERSED MICELLE OF DIPALMITOYLPHOSPHATIDYLCHOLINE

Abstract:
One and two-dimensional nuclear magnetic resonance studies at 300 MHz have been performed on gramicidin S in DPPC reversed micelle.The conventional method of sequential assignments involving J-correlated spectroscopy (COSY) and nuclear Overhauser spectroscopy (NOESY) was used.All resonance peaks of gramicidin S were assigned.And then,the NMR assignments were used to determine the three dimensional structure of gramicidin S in DPPC reversed micelle.Ratios of interproton distances were obtained from NOESY spectrum,and HN-CaH coupling constants of Leu and Orn residues derivated from spin echo spectrum were used to calibrate the interproton distances.The results indicate that gramicidin S bendsat Ca of the two ornthine residues when it is bound to revetsed micelle of DPPC.
