Protoplast Isolation from Root Suspension Culture Cells and Plant Regeneration in Rice

Abstract:
After loose calli are produced from tender roots Oryza sativa L. on MS and N6 media, much active protoplasts are obtained by means of protoplasts isolated from callus-derived shock suspension culture in an agitated liquid AA medium. The experiment shows that this is an ideal material to isolate protoplasts . Calli from protoplasts are developed in MS agarose medium containing 2, 4-D Img/L (the following unit is the same ), KT 0. 2, NAA 0. 2, glutamine 867; asparagine 266. Plantlets are induced on N6 medium containing KT 2, NAA 0. 2, CH 1000, 4% coconut milk. The plants generated from protoplast R! generation are able to bloom, produce and seed normally; the number of chromosomes is diploidy (2n = 24). The most characteristic of all is the low rate of the seed setting, reduction in the number grains of each spike and the prolonged time of growth .
