Molecular detection of symbiotic bacterium Buchera from aposymbiotic aphid Sitobion avenae

Abstract:
To detect the aposymbiotic effect of aphids is of significance in understanding the symbiotic system of aphids and Buchera. In this paper, the aposymbiotic effect of Sitobion avenae on symbiotic bacterium Buchera was studied by PCR technique combined with transmission microscope. The results showed that after treated S. avenae with rifampicin for 5- and 10-days, some expected fragments in Buchera bacteria were detected, suggesting that the symbionts in aphid body cavity were not injured. However, after treated with rifampicin for 15-days, the fragments disappeared, suggesting that the symbionts were completely disrupted. The observation with transmission microscopy was consistent with the diagnosis with PCR.
