MICRO CARBON FIBER ELECTRODE FOR RECORDING OF QUANTAL SECRETION IN SINGLE CELL

Abstract:
Biophysical technologies for single cell recording was described, with special focus on micro carbon fiber electrode (CFE) and its application in stimulus-secretion coupling. In contrast to membrane capacitance of patch-clamp recording, CFE has advantages in high sensitivity, real detection of release substance and no need for tight-seal patch-clamp, although it detects only a few easily oxidizable transmitters including catecholamines and serotonin. In combination with patch-clamping and Ca imaging measurement, coupling between action-potentials and quantal transmitter secretion in chromaffin cells are demonstrated as the example of CFE application.
