THE RECEPTOR AGONIST-LIKE EFFECTS OF SAIKOSAPONIN(I) IN ISOLATED RAT PANCREATIC ACINAR CELLS

Abstract:
The effects of Bt2-cGMP and GDP on Saikosaponin and CCK-8 stimulated amylase secretion and i increase were investigated in isolated rat pancreatic acinar cells. Bt2-cGMP inhibited both SA(I) and CCK-8-induced amylase secretions in a dose-dependent way. GDP also inhibited markedly saikosaponin-induced amylase secretion. SA(I) induced i increase in single pancreatic acinar cell was different from that induced by CCK-8: the rising phase of i increase was slow to develop, and lasted longer. In addition, there was a secondary increase after the initial peak. GDP significantly inhibited SA(I)-induces initial peak of i increase. These data indicate that SA(I) may activate i increase and amylase secretion via a direct action on the cell surface receptor.
