Abstract: OBJECTIVE: To scrutinize the protective role of hydroethanolic extract of Euphorbia neriifolia leaves (HEEN) against N-nitrosodiethylamine (DENA)-induced hepatocarcinogenesis in male Swiss albino mice. METHODS: Experimental mice were pretreated with 150 and 400 mg/kg of HEEN, or 0.5% and 1% of butylated hydroxyanisole (BHA) as a standard for 14 d prior to the administration of a single dose of 50 mg/kg of DENA. Levels of xenobiotic metabolizing enzymes such as cytochrome (Cyt) P450 and Cyt b5, activities of reduced glutathione (GSH), glutathione-S-transferase (GST), aspartate aminotransaminase (AST), alanine aminotransaminase (ALT) and alkaline phosphatase (ALP), and total protein and cholesterol content in the liver tissues were measured to determine the hepatotoxicity caused by DENA.RESULTS: The levels of Cyt P450 and Cyt b5 were significantly increased, and GST and GSH were significantly depleted after DENA treatment (P<0.01). The activities of AST, ALT and ALP, and the total protein content were also significantly dropped off (P<0.01). The total cholesterol level was markedly increased by DENA as compared with the normal group (P<0.01). However, the pre-supplementation of HEEN showed a remarkable amelioration of these abnormalities, and the levels of the antioxidant enzymes in the liver were significantly restored, which exhibited the dose-dependent protective effect against DENA-induced hepatotoxicity. CONCLUSION: HEEN exerts its chemopreventive effects by alleviating the xenbiotic enzymes and enhancing the levels of antioxidants and biochemical assays in DENA-induced carcinogenesis by reducing the formation of free radicals.

Abstract: The antioxidant activity of the methanol extract of leaves of Hyptis suaveolens Poit. was evaluated in vitro by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity using gallic acid and butylated hydroxyanisole (BHA) as reference standards. They exhibited strong antioxidant radical scavenging activity with IC50 value of 0.4, 1.15 and 14.04 μg mL-1 for Gallic acid, BHA and Hyptis suaveolens, respectively. The antioxidant activity of methanol extract could be due to the presence of Flavonoids.

Abstract: A new method for the determination of butylated hydroxyanisole (BHA) and butylated hydroxytoluene (BHT) in cosmetics by gas chromatography-mass spectrometry selected ion storage (SIS) method was developed. The BHA and BHT in samples were extracted by methanol. The m/z 165 and m/z 205 were the monitoring ion for BHA and BHT respectively. The detection limits of BHA and BHT in samples were 2.5 micrograms/g and 0.5 microgram/g, respectively. The method is simple, rapid and accurate.

Abstract: In model experiments with the use of Miller's test to 5 ml emulsion 25, 12.5, 6.25 and 3.13μg BHA, BHT and TBHQ was added. Inhibitory ratios (IR) for BHA and BHT were similar. At addition level of 6.25 and 3.13 IR for TBHQ was lower than for BHA and BHT. It is suggested that in the model studies with the use of Miller's test 25 μg BHA, BHT or TBHQ should be added to 5 ml of control sample. En experimentos modelos usando el test de Miller, a 5ml de emulsión se a adió 25, 12.5, 6.25 y 3.13μg de BHA, BHT y TBHQ. Las relaciones inhibitorias (IR) para el BHA y BHT fueron similares. IR para TBHQ fue más baja que para BHA y BHT con niveles de adición del 6.25 y 3.13. Esto sugiere que en estudios modelos usando el test de Miller 25μg de BHA, BHT, o TBHQ deben ser a adidos a 5 ml de muestra control.
Rational use of antioxidants in solid oral pharmaceutical preparations

Abstract: Antioxidants are currently used as efficient excipients that delay or inhibit the oxidation process of molecules. Excipients are often associated with adverse reactions. Stability studies can guide the search for solutions that minimize or delay the processes of degradation. The ability to predict oxidation reactions in different drugs is important. Methods: This study was conducted to assess the rational use of butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), sodium metabisulfite (SMB), propyl gallate (PG) and cysteine (CYS) in tablet formulations of simvastatin and ketoconazole. These antioxidants were evaluated according to stability parameters and the relationship between efficiency of the antioxidant and chemical structure of the drugs. Results were compared with DPPH tests and computational simulations. BHT was most efficient regarding simvastatin stability, and the most effective BHT concentrations for maintaining stability were 0.5 and 0.1%. In relation to ketoconazole, SMB was most efficient for maintaining content and dissolution profile. The evaluation by DPPH showed that the largest percentage of absorbance reduction was observed for PG, while SMB proved most efficient and had lower consumption of DPPH. The same pattern was observed, albeit with lower efficiency, for the other lipophilic antioxidants such as BHT and BHA. The results of the molecular modeling study demonstrated that electronic properties obtained were correlated with antioxidant activity in solution, being useful for the rational development of liquid pharmaceutical formulations but not for solid oral formulations. This study demonstrated the importance of considering stability parameters and molecular modeling to elucidate the chemical phenomena involved in antioxidant activity, being useful for the rational use of antioxidants in the development of pharmaceutical formulations. Abstract: there are several factors that can lead to the instability of an emulsion, highlighting the oxidation, a reaction prevented by the use of antioxidants. the butylated hydroxytoluene (bht) has been one of the most used synthetic antioxidants in cosmetic formulations; however, pharmaceutical and cosmetic industries have shown considerable interest regarding the search for the use of natural products. based on this, the objective of this work was the incorporation of resveratrol, a natural phenolic compound found mainly in grapes as well as in red wines, into a non-ionic emulsion basis for assessing the profile of stability and antioxidant activity, as compared with a non-ionic basis emulsion containing bht. the profile of stability was examined by the observation of the organoleptic characteristics, determination of ph and spreadability, and the antioxidant activity through the radical scavenging dpph test. the results showed that the emulsion containing bht was more stable than the emulsion containing resveratrol, when high temperature was used. for the analysis of the antioxidant activity, the resveratrol, in both forms of incorporation, showed significant antioxidant activity in comparison to bht, suggesting that resveratrol may be a viable antioxidant alternative to be used into cosmetic preparations.

Abstract: The implications of diet on health sustainability have assumed a major importance. Microcystis are able to enhance the nutritional content of conventional food and feed preparation and hence to positively affect humans and animal health due to their original chemical compositions, protein contents, antioxidant properties, vitamins and other biologically active compounds. Butylated hydroxytoluene ‘BHT’ is one of the synthetic antioxidant agents commonly used for food additives. Cyanobactria like Oscillatoria sp. and Microcystis aerungionosa, those are commonly available in the coastal zone of Puri District of Odisha State were capable of producing this compound. Extracts from these two species exhibited various degrees of antioxidant properties when they were tested with free radical scavenging assay. The highest antioxidant activity was observed in the extracts of Microcystic aerungionosa, which displayed a similar activity to synthetic BHT. Gas chromatography and mass spectroscopy analysis concluded that the compounds are very similar to the synthetic BHT. This synthetic antioxidant was produced in the cells irradiated with higher light intensity. More BHT was produced in the cells irradiated with a higher light intensity and its production was irradiance dependent. The quantity of cellular BHT displayed a positive correlation with antioxidant activity of the tested species. The present study confirms the production of natural antioxidant like BHT from these two species of Puri District of Odisha State which constitute a potential source for producing natural antioxidant.
