An Integrated Approach Based on Multiplexed Protein Array and iTRAQ Labeling for In-Depth Identification of Pathways Associated to IVF Outcome

Abstract: The emergence of high-throughput protein quantification methodologies has enabled the comprehensive characterization by longitudinal and cross-sectional studies of biological fluids under physiological and pathological conditions. In particular, the simultaneous investigation of cytokines and growth factors signaling pathways and their associated downstream effectors by integrated multiplexed approaches offers a powerful strategy to gain insights into biological networks and processes in living systems. A growing body of research indicates that bioactive molecules of human reproductive fluids, including human follicular fluid (hFF), may affect oocyte quality, fertilization and embryo development, thus potentially influencing the physiopathology of pregnancy-related conditions. In this work, an iTRAQ labeling strategy has been complemented with a multiplexed protein array approach to analyze hFFs with the aim to investigate biological processes and pathways related to in vitro fertilization (IVF) outcome. The iTRAQ labeling strategy lead to the quantification of 89 proteins, 30 of which were differentially expressed in hFFs with successful compared to unsuccessful IVF outcome. The targeted study, based on multiplexed antibody protein arrays, allowed the simultaneous quantification of 27 low abundance proteins, including growth factors, chemokines and cytokines endowed with pro- and anti-inflammatory activity. A significant number of differentially regulated proteins were involved in biological functions related to blood coagulation, acute phase response signaling and complement system. Overall, the present results provide an integrated overview of protein changes in hFFs associated to IVF outcome, thus improving current knowledge in reproductive medicine and fertility research.
Integrated Scenario Planning and Multi-Criteria Decision Analysis Framework with Application to Forest Planning  [PDF]

Abstract: This paper explores approaches concerning complex forest planning challenges, such as restoration after large-scale disturbances and under climate change. It introduces a new framework that integrates qualitative scenario planning with quantitative multi-criteria decision analysis. This framework allows stakeholders without background in forestry to express their preferences as a set of scenarios that are further assessed for specific forest management goals and activities using multi-criteria models. The assessment of the modelled scenarios created a common understanding for the stakeholders and experts to compare trade-offs between several management options and needed policy choices. The framework was applied in the case study of forest restoration following insect disturbance in British Columbia, Canada. The framework enabled structured stakeholder groups’ interactions such as industry, business associations, local and regional governments, and non-governmental organizations to identify potential restoration options. Different community futures were envisioned by two scenarios: one resembling current conditions and standard practices, while another promoting diversification of the forestry sector. The results indicated that each of the scenarios leads to different consequences for the community measured by levels of economic benefits, total harvest volumes and harvest flows over time. The results also show that the developed framework linking scenarios and multi-criteria decision analyses proved crucial to broaden the discussion on relevant species mixes and management practices, and their implications for the community and policy development.

Abstract: Exploratory studies developed at several neurosciences laboratories at universities around the world show us through the experience that there is a biological process called neuroplasticity. Because of this oldest concept about the neuronal formation, scientists also thought that if a particular area of the adult brain was damaged, the nerve cells could not form new connections and the functions controlled by this field of the brain would be permanently lost or could not be regenerate. However, studies have overturned this old view, and currently, scientists recognize that the brain continues to reorganize itself by forming new neural connections during the life. This phenomenon is called neuroplasticity that refers to the potential which the brain should be reorganized by creating new neural pathways to adapt, as it needs.

A new method to compute a global view of a scene is presented in the paper. The global view of a scene is determined by a “good” set off points of view. This method is based on a genetic algorithm. The “good” set of points of view is used to compute a camera path around the scene.

Abstract: the main purpose of this article is to explore the relationship between food and sociability. although this is a theme often approached in anthropology, the true role of food in this relationship is seldom made explicit. based on three different empirical researches, the article identifies the kind of sociability that is established in each meal in accordance with the day of the week, indicating for each case the degree of relevance assumed by food itself. following the daily sequence of the different meals - breakfast, lunch and dinner -, the data presented allows for the discussion of the definition of sociability as developed by simmel and its application to the context of brazilian society.

Abstract: The purpose of this article is to analyze from the perspective of the game-theoretical lenses the most adequate way(s) to interact for two actors: the European Union and the Black Sea region (seen as unitary player and at individual states’ level). An important element lies in the fact that the region is not homogenous, the countries demonstrating different levels of economic development. The pillars where a country needs to undertake specific measures, in order to determine progress for its economic development, are highlighted in the Global Competitiveness Report. These pillars include, ultimately, the various domains of interaction and form a structure for the international cooperation. The strategy of cooperation that an international actor chooses must bring an advantage (or a concrete gain value) and basically ensure its own economic growth and increased competitiveness with pre-requisites allowing the advancement to a superior economic development stage.
