Statistical Analysis of fMRI Time-Series: A Critical Review of the GLM Approach

Abstract: Functional magnetic resonance imaging (fMRI) is one of the most widely used tools to study the neural underpinnings of human cognition. Standard analysis of fMRI data relies on a general linear model (GLM) approach to separate stimulus induced signals from noise. Crucially, this approach relies on a number of assumptions about the data which, for inferences to be valid, must be met. The current paper reviews the GLM approach to analysis of fMRI time-series, focusing in particular on the degree to which such data abides by the assumptions of the GLM framework, and on the methods that have been developed to correct for any violation of those assumptions. Rather than biasing estimates of effect size, the major consequence of non-conformity to the assumptions is to introduce bias into estimates of the variance, thus affecting test statistics, power, and false positive rates. Furthermore, this bias can have pervasive effects on both individual subject and group-level statistics, potentially yielding qualitatively different results across replications, especially after the thresholding procedures commonly used for inference-making.

Abstract:

Interpreting oxygenation-based neuroimaging signals: the importance and the challenge of understanding brain oxygen metabolism

Abstract: Functional magnetic resonance imaging is widely used to map patterns of brain activation based on blood oxygenation level dependent (BOLD) signal changes associated with changes in neural activity. However, because oxygenation changes depend on the relative changes in cerebral blood flow (CBF) and cerebral metabolic rate of oxygen (CMRO2), a quantitative interpretation of BOLD signals, and also other functional neuroimaging signals related to blood or tissue oxygenation, is fundamentally limited until we better understand brain oxygen metabolism and how it is related to blood flow. However, the positive side of the complexity of oxygenation signals is that when combined with dynamic CBF measurements they potentially provide the best tool currently available for investigating the dynamics of CMRO2. This review focuses on the problem of interpreting oxygenation-based signals, the challenges involved in measuring CMRO2 in general, and what is needed to put oxygenation-based estimates of CMRO2 on a firm foundation. The importance of developing a solid theoretical framework is emphasized, both as an essential tool for analyzing oxygenation-based multimodal measurements, and also potentially as a way to better understand the physiological phenomena themselves. The existing data, integrated within a simple theoretical framework of O2 transport, suggests the hypothesis that an important functional role of the mismatch of CBF and CMRO2 changes with neural activation is to prevent a fall of tissue pO2. Future directions for better understanding brain oxygen metabolism are discussed.

Abstract: Blood oxygen level-dependent magnetic resonance imaging (BOLD MRI) is a noninvasive technique useful in patients with renal disease. The current study was performed to determine whether BOLD MRI can contribute to the diagnosis of renal pathological patterns. BOLD MRI was used to obtain functional magnetic resonance parameter R2* values. Gray-level co-occurrence matrixes (GLCMs) were generated for gray-scale maps. Several GLCM parameters were calculated and used to construct algorithmic models for renal pathological patterns. Histopathology and BOLD MRI were used to examine 12 patients. Two GLCM parameters, including correlation and energy, revealed differences among four groups of renal pathological patterns. Four Fisher’s linear discriminant formulas were constructed using two variables, including the correlation at 45° and correlation at 90°. A cross-validation test showed that the formulas correctly predicted 28 of 36 samples, and the rate of correct prediction was 77.8%. Differences in the texture characteristics of BOLD MRI in patients with lupus nephritis may be detected by GLCM analysis. Discriminant formulas constructed using GLCM parameters may facilitate prediction of renal pathological patterns

Abstract:
目的:探讨伴有认知功能改变的脑震荡急性期患者功能磁共振低频振幅(ALFF)变化与其临床症状之间的关系。方法:采用静息态血氧水平依赖功能磁共振成像(BOLD-fMRI)及其低频振幅(ALFF)计算法,对急性期伴有认知功能改变的脑震荡患者在静息状态下获得相关资料进行分析。研究组为脑震荡成年患者10人;对照组为健康成年志愿者10人。研究组10人均在伤后72小时内完成BOLD-fMRI检查。两组均采用GE公司制造MR750 3.0T磁共振成像系统,在静息态下采集BOLD-fMRI成像数据。结果:与对照组比较,研究组在边缘叶、扣带回、楔前叶、左侧颞上回、颞中回、右侧额上回等脑区表现为ALFF值升高,临床上出现记忆能力、语言表达、情绪反应等认知功能改变。结论:脑震荡急性期患者认知功能异常,可能与边缘叶、扣带回、右侧前额叶等脑区ALFF值升高相关。
Objective: To investigate the change of amplitude of low-frequency fluctuation (ALFF) in acute phase patients with concussion. Methods: 10 patients with concussion and 10 healthy control subjects were performed at GE MR750 3T by using the methods of blood oxygenation level de-pendent-functional magnetic resonance imaging (BOLD fMRI) and ALFF in this study. Results: Compared with the control subjects, the patients with concussion showed significantly increased ALFF values in limbic lobe, cingulate gyrus, precuneus, left superior temporal gyrus, left middle temporal gyrus and right frontal gyrus. The clinical manifestations of these patients were the changes of cognitive function such as memory, language and emotional responses. Conclusion: The changes of cognitive function on the patients with concussion were associated with the increased ALFF values in the cognitive-related brain areas.
