芳香烃受体在干眼中的调控作用

Abstract:
目的:探讨芳香烃受体(Aryl hydrocarbon receptor, AhR)在干眼中对结膜杯状细胞增殖分化及黏蛋白5AC (MUC5AC)分泌的调控作用。方法:建立小鼠干眼动物模型,采用酚红棉线法测量小鼠泪液分泌量,使用裂隙灯观察小鼠角膜荧光素钠染色损伤情况,并记录临床评分。给予AhR激动剂(FICZ)或AhR抑制剂(CH223191)处理七天,采用RT-PCR检测小鼠结膜组织中AhR、MUC5AC和炎症因子(TNF-α, IL-1β, IL-6, IL-8) mRNA的表达。提取C57BL/6小鼠结膜杯状细胞,给予FICZ或CH223191处理,采用RT-PCR检测小鼠结膜杯状细胞中AhR、MUC5AC和炎症因子(TNF-α, IL-1β, IL-6, IL-8) mRNA的表达。结果:与对照组相比,AhR在干眼小鼠结膜组织中表达降低。FICZ处理后AhR、MUC5AC表达升高,炎症因子表达下降。CH223191处理后AhR、MUC5AC表达降低,炎症因子表达升高。结论:AhR活化后促进结膜杯状细胞增殖分化,增加MUC5AC分泌,在治疗干眼中发挥重要作用。
Objective: To investigate the effect of Aryl hydrocarbon receptor (AhR) on the proliferation and dif-ferentiation of conjunctival goblet cells and the secretion of mucin 5AC (MUC5AC) in dry eye. Meth-ods: Dry eye models of mice were developed to measure the tear volume by using phenolic cotton line method, observe the corneal injury stained with sodium fluorescein under a slit-lamp, and rec-ord the clinical scores. RT-PCR was used to detect the mRNA expressions of AhR, MUC5AC and in-flammatory cytokines (TNF-α, IL-1β, IL-6, IL-8) in conjunctiva tissues treated by the AhR agonist (FICZ) or the AhR inhibitor (CH223191) for 7 days. The conjunctiva goblet cells of C57BL/6 mice were collected and treated with FICZ or CH223191. RT-PCR was used to detect the mRNA expres-sions of AhR, MUC5AC and inflammatory cytokines (TNF-α, IL-1β, IL-6, IL-8) in conjunctiva goblet cells of mice. Results: Compared with the control group, the expression of AhR was decreased in the conjunctiva tissue of dry eye mice. The expressions of AhR and MUC5AC were increased by FICZ, and the expressions of inflammatory factors were decreased by FICZ. The expressions of AhR and MUC5AC were decreased by CH223191, and the expressions of inflammatory factors were increased by CH223191. Conclusion: The activation of AhR plays an important role in the treatment of dry eye by promoting the proliferation and differentiation of conjunctival goblet cells and increasing the secretion of MUC5AC.
