延胡索总生物碱微透析探针体外回收率测定研究

Abstract:

目的:建立延胡索甲素、延胡索乙素、D-四氢药根碱、脱氢紫堇碱为标准的延胡索总生物碱经皮给药微透析采样的体外回收率测定方法。方法:运用超高液相色谱法采用浓度差法(增量法,减量法)测定其体外回收率,并考察灌流液的pH、流速、浓度、温度对回收率的影响,以探讨微透析技术用于延胡索生物碱体内药动学研究的可行性。结果表明,增量法和减量法所测得的探针体外回收率在相同的条件下近似相等;相同的条件下,探针体外回收率在流速为0.5~1.5 μL/min范围内随着流速增加而降低,在温度为25℃~42℃范围内随着温度上升而明显增加;同一流速下,探针体外回收率与延胡索乙素浓度无关。结论:建立了延胡索总碱经皮给药的微透析采样方法,最终选择了pH 4,流速为0.5 μL/min,取样时间间隔为80 min,温度为37℃,可用于延胡索总碱给药后皮肤药动学研究。
Objective: To establish a method for determination of recovery rate of total alkaloids of corydalis fumaricum, fumaricum ethyl, D-tetrahydrorhizine and dehydrocorydaline by microdialysis sam-pling. Methods: The in vitro recovery rate was measured by the concentration difference method (increment method, declination method) by ultra high liquid chromatography. The effects of pH, velocity, concentration and temperature of the perfusion solution on the recovery rate were inves-tigated, so as to explore the feasibility of microdialysis technology for pharmacokinetic study of co-rydalis corydalis alkaloids in vivo. The results show that the recoveries of the probes measured by increment method and decrement method are approximately equal under the same conditions. Under the same conditions, the recovery rate of probes decreased with the increase of flow rate in the range of 0.5~1.5 μL/min, and increased with the increase of temperature in the range of 25~42?C. At the same flow rate, the recovery rate of probe was not related to the concentration of rhizoma fumaricum ethyl. Conclusion: A microdialysis sampling method was established for percu-taneous administration of total rhizoma corydalis. The pH 4, the flow rate of 0.5 μL/min, the sam-pling interval of 80 min and the temperature of 37?C were selected, which can be used for the study of the pharmacokinetics of the skin after the administration of total rhizoma corydalis.
