Biodiversity Of Aspergillus Species Isolated From Sogutluk Forest Soil Of Edirne City

We aimed to isolation and identification of Aspergillus species in Edirne Sogutluk Forest soil by morphological-colonial and molecular methods in our study. For this purpose, soil samples were taken by using Brown's Method in March, 2016 from mentioned forest soil. “Soil Dilution Method” was used for fungus isolation from soil. For the morphological studies, microscopic and macroscopic characters were examined after the operation of inoculating in CZ, CYA, CY20S, MEA media. For the molecular studies, DNA isolation, PCR operations that targeting Calmodulin gene region, purification of the PCR product, sequence analysis and phylogenetics analysis processes were performed respectively. Seven Aspergillus species were detected as a result of studies. These species are Aspergillus affinis (New record for Turkey), Aspergillus awamori, Aspergillus carbonarius, Aspergillus dimorphicus, Aspergillus europaeus (New record for Turkey), Aspergillus spelaeus (New record for Turkey) and Aspergillus fischeri
