The recently solved structure of the Shh receptor Ptch may provide insight into how Disp binds and releases Hh family ligands. Disp is activated by Furin-mediated processing. The secreted glycoprotein Scube2 collaborates with Disp during Shh deployment. Disp directs endosomal recycling of Hh to facilitate release of the long-range signal. Disp localizes to and can modulate the activity of cytonemes to affect Hh delivery to target cells
