摘要 目的 探讨IL-6、细菌脂多糖(LPS)激活巨噬细胞所得上清液(CM)对神经母细胞瘤细胞株Neuro2a细胞增殖、凋亡及细胞突起生长与Sez6基因表达的影响。 方法 以IL-6和CM分别作用于Neuro2a细胞,采用MTT法、流式细胞术分别检测细胞的增殖、凋亡能力;培养24 h后观察统计细胞突起生长变化,采用实时荧光定量PCR(RT-qPCR)方法检测Sez6基因的表达。 结果 与对照组比较,IL-6、CM刺激对Neuro2a细胞增殖、凋亡未见明显影响,但可增加细胞突起总长度、最长突起长度、突起数目,并且可促进Sez6基因的表达(F=6.00~22.52,P<0.05)。 结论 IL-6、CM对Neuro2a细胞增殖、凋亡作用不明显,但可能通过上调Sez6基因的表达促进细胞突起的生长。 Abstract:Objective To investigate the effects of interleukin-6 (IL-6) and the supernatant of lipopolysaccharide (LPS)-activated macrophages on the proliferation, apoptosis, neurite outgrowth, and Sez6 gene expression of neuroblastoma Neuro2a cells. Methods Neuro2a cells were treated with IL-6 and conditioned medium (CM) collected from the supernatant of LPS-activated macrophages. MTT assay and flow cytometry were performed to analyze cell proliferation and apoptosis. After 24 h of incubation, the change in neurite outgrowth was observed. Quantitative real-time PCR was performed to measure the mRNA expression of Sez6. Results Compared with the control group, stimulation with IL-6 or CM had no significant influence on the proliferation and apoptosis of Neuro2a cells, but significantly increased the total length of neurites, the longest length of neurites, and the number of neurites as well as the mRNA expression of Sez6 (F=6.00-22.52,P<0.05). Conclusion IL-6 and CM may not significantly affect the proliferation and apoptosis of Neuro2a cells, but they could promote neurite outgrowth by upregulating the mRNA expression of Sez6
