Abstract:

摘要:目的 研究苯并咪唑出芽抑制解除同源物蛋白1(budding uninhibited by benzimidazoles 1, BUB1)对胶质母细胞瘤(glioblastoma, GBM)细胞增殖的促进作用。方法 利用TCGA数据库进行生物信息学分析及Real-time PCR和Western blot等分子生物学技术对GBM细胞中BUB1的表达水平进行检测,并利用慢病毒转染siRNA沉默BUB1的表达以探究其对于GBM细胞增殖能力的影响;同时,利用生物信息学手段对Rembrandt数据库的预后资料进行分析,探究BUB1与GBM患者预后生存的相关性。结果 BUB1在GBM中高表达,而特异性沉默BUB1能够显著抑制U87细胞的生长(P=0.007)和克隆形成的能力(P=0.033);BUB1低表达组GBM患者的临床预后显著优于高表达组患者(P=0.040)。结论 BUB1表达上调能够促进GBM细胞的生长和增殖,并和GBM患者的不良预后相关。
ABSTRACT: Objective To investigate the functions of mitosiskinase budding uninhibited by benzimidazoles 1 (BUB1) for cell proliferation in glioblastoma (GBM). Methods We investigated the expression level of BUB1 in GBM cells by analyzing TCGA database. Then Real-time PCR and Western blot were used to explore the role of BUB1 in proliferation of GBM cells via BUB1 silencing using siRNA transfection. Additionally, we investigated the correlation between BUB1 expression and prognosis of GBM patients. Results BUB1 expression was significantly elevated in GBM patients, while artificial silencing of BUB1 attenuated the proliferation (P=0.007) and cloning formation of U87 cells (P=0.033). At the same time, improved prognosis could be observed in GBM patients with highly expressed BUB1 compared with those with lower BUB1 expression (P=0.040). Conclusion Increased BUB1 expression promotes the growth and proliferation of GBM cells and is correlated with the poor prognosis of GBM patients
