LINC00337促进肺腺癌细胞增殖

Abstract:

近年来,长非编码RNAs(long non-coding RNAs, lncRNAs) 在肺腺癌发生发展中的作用备受关注。生物信息学预测,长非编码RNA337(LINC00337)在肺腺癌组织中的表达量显著高于其在正常组织中的表达量,且其高表达与患者较差的预后正相关。qRT-PCR结果发现,LINC00337在肺腺癌细胞中的表达量也高于正常肺上皮细胞:LINC00337在肺腺癌细胞H1975和A549中的表达量分别是其在正常肺上皮细胞BEAS-2B中的2.6和10.3倍。在肺腺癌细胞中,上调LINC00337将增加细胞周期蛋白E2 (cyclin E2, CCNE2)的表达,进而促进该细胞增殖;相反,敲低LINC00337将抑制CCNE2的表达,最终阻止肺腺癌细胞增殖。生物信息学预测结合染色质沉淀实验证实,信号转导和活化转录因子1(signal transducer and activator of transcription 1, STAT1)可结合在CCNE2启动子上,下调其表达。在H1975细胞中,过表达STAT1可下调57.76%的CCNE2。在此细胞中,同时过表达STAT1和敲低LINC00337,发现该组细胞的CCNE2表达量增加1.62倍。相反,在A549细胞中敲低STAT1,发现CCNE2可增加6.48倍表达量。但在此细胞中,同时敲低STAT1和过表达LINC00337,则发现该组细胞的CCNE2表达量无变化。上述结果提示,LINC00337对CCNE2的调控需要STAT1的存在。总之,LINC00337通过转录因子STAT1调控CCNE2表达促进肺腺癌细胞增殖
