吉非替尼联合鸦胆子油乳对肺腺癌细胞的作用及其机制

Abstract:
摘要 目的 探讨吉非替尼（Gefitinib, G）联合鸦胆子油乳（Bruceolic oil emulsion, B）对肺腺癌PC-9、A549细胞增殖与凋亡的作用及其机制。方法 取对数生长期的PC-9、A549细胞进行实验，两细胞株在实验中均分为对照组和实验组。采用MTT比色法、TUNEL法及流式细胞技术检测各实验组对肺腺癌细胞的增殖抑制、诱导细胞凋亡及细胞周期阻滞等作用。结果 吉非替尼、鸦胆子油乳单药均能抑制PC-9、A549细胞的生长、增殖并诱导其凋亡，呈浓度和时间依赖效应。与鸦胆子油乳联合后，吉非替尼对PC-9和A549细胞的IC50较单用时分别下降了50.72%和66.56%。两药联合后对两细胞的增殖抑制及凋亡诱导作用进一步增强。结论 吉非替尼联合鸦胆子油乳能显著抑制肺腺癌PC-9、A549细胞增殖、诱导细胞凋亡，可能与药物对细胞周期的阻滞作用有关。两者合用时鸦胆子油乳可对吉非替尼起到增效增敏的作用
