Abstract:
摘要 目的 探讨FoxO3在膀胱癌中的表达及其与患者预后和临床病理特征的相关性。方法 收集111例膀胱癌患者的癌组织及癌旁正常组织标本，采用实时定量PCR、免疫组织化学方法和Westernblot法检测FoxO3的表达。分析FoxO3与膀胱癌临床病理特征的相关性。对FoxO3进行Kaplan-Meier法生存分析。采用Cox比例风险模型分析影响预后的独立因素。结果 FoxO3基因的mRNA和蛋白在膀胱癌组织中的表达均低于癌旁正常组织，差异有统计学意义（P<0.05）。膀胱癌淋巴结转移组的FoxO3蛋白的阳性率显著低于无淋巴结转移组（P<0.05）。TNM分期中Tis~T1浅表性膀胱癌组FoxO3蛋白的阳性率显著低于T2~T3浸润性膀胱癌组。组织学分型中高级别组的FoxO3蛋白阳性率显著低于低级别组（P<0.05）。膀胱癌组织中FoxO3蛋白表达与疾病的恶化程度呈正相关（P<0.05）。FoxO3蛋白表达阴性、淋巴结转移是膀胱癌患者预后不良的危险因素。结论 FoxO3蛋白表达在膀胱癌组织中显著低表达，其阴性表达是膀胱癌患者预后不良的危险因素
